Mechanism of action

GHS-R1a Agonism & GH Secretion

Ipamorelin selectively activates GHS-R1a on somatotrophs, coupling via Gฮฑq/11 to PLCฮฒ โ†’ IPโ‚ƒ/DAG signaling, triggering Caยฒโบ-dependent exocytosis of growth hormoneโ€”yielding pulsatile release without concomitant ACTH or cortisol secretion.

Anabolic & Lipolytic Signaling

Elevated GH induces hepatic IGF-1 production, which engages the IGF-1R/PI3K/Akt pathway in muscle and adipose tissue, promoting protein synthesis and adipocyte lipolysis.

Osteoanabolic Effects

GH/IGF-1 axis activation upregulates Runx2 and osterix in osteoblasts via JAK2/STAT5 signaling, enhancing bone matrix deposition and mineralization.
